DSIP (Delta Sleep-inducing Peptide) is a neuropeptide that, when delivered into the mesodiencephalic ventricle of recipient rabbits, induces spindle and delta EEG activity along with reduced motor activity. Numerous roles for DSIP have been proposed through research using peptide analogues with greater molecular stability and by measuring DSIP-like immunoreactivity (DSIP-LI) responses after administration of DSIP antiserum and antibodies. It is a well-known neuromodulator and natural sleep-promoting nonapeptide with many additional physiological functions. Typically found in the brain, it readily crosses the blood-brain barrier. It is believed to enhance sleep regulation, reduce motor activity to support deep sleep, stimulate LH levels, and reportedly help normalize blood pressure. DSIP can also alter serum levels of certain hormones, including adrenocorticotropic hormone, somatostatin and serotonin. It is an amphiphilic peptide with a molecular weight of approximately 850 daltons and the amino acid sequence N-Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu-C. It has been detected in both free and bound forms in the hypothalamus, limbic system and pituitary, as well as in various peripheral organs, tissues and body fluids.

Application

In the pituitary, DSIP co-localizes with numerous peptide and non-peptide mediators, including corticotropin-like intermediate peptide (CLIP), adrenocorticotropic hormone, melanocyte-stimulating hormone (MSH), thyroid-stimulating hormone (TSH) and melanin-concentrating hormone (MCH). Delta-sleep-inducing peptide is also abundant in gut secretory cells and in the pancreas, where it co-localizes with glucagon. DSIP is believed to promote sleep regulation, reduce motor activity to support deep sleep, stimulate LH levels, and reportedly help normalize blood pressure. In addition, DSIP can influence the serum levels of certain hormones, such as adrenocorticotropic hormone, somatostatin and serotonin.
