5-Isoquinolinamine,3-chloro-(9CI) Use and Manufacturing

To a solution of 3-chloro-5-nitroisoquinoline (2.3 g, 1 1.0 mmol) in acetic acid (100 mL) was added zinc dust (3.5 g, 55.1 mmol) in portions. The resulting mixture was stirred for 2 hours at ambient temperature. Then the resulting mixture was filtered through Celite and the filtrate was concentrated under reduced pressure. The residue was purified by silica gel column chromatography, eluted with 1percent methanol in dichloromethane to afford 3-chloroisoquinolin-5-amine as an off-white solid: MS (ESI, m/z): 179.0 [M + 1]A solution of Example 81B (10 mg, 0.048 mmol) in tetrahydrofuran (2 mL) was added to water-wet Ra-Ni (30.0 mg, 0.511 mmol) and exposed to hydrogen gas at 30 psi.
