6-Fluoro-2-methyl-3-pyridinecarboxylic acid Use and Manufacturing

6-Fluoro-2-methylnicotinic acid (400 mg, 2.58 mmol) and DIPEA (900 muL, 5.16 mmol) were added to a solution of (R)-3-[l-(3-isopropyl-[l, 2, 4]oxadiazol-5-yl)piperidin-4-yl]- butylamine (Preparation 14, 690 mg, 2.58 mmol) in NMP (3 mL) and the resulting solution heated in the microwave at 1000C for 7 h. The reaction mixture was diluted with EtOAc, washed with IM HCl and the acidic washings basified to pH 5 with IM NaOH, before being extracted with EtOAc. The organic extracts were washed with H2O, brine, dried (MgSO4), filtered and concentrated in vacuo. Purification by column chromatography (EtOAc-IH-AcOH; 69:30:1) afforded the title compound: RT = 2.70 min; mlz (ES+) = 402.69 [M + H]+.
