Description

ChEBI: Peramivir is a member of the class of guanidines that is used (as its trihydrate) for the treatment of acute uncomplicated influenza in patients 18 years and older who have been symptomatic for no more than two days. It has a role as an antiviral drug and an EC 3.2.1.18 (exo-alpha-sialidase) inhibitor. It is a member of cyclopentanols, a member of acetamides, a member of guanidines and a 3-hydroxy monocarboxylic acid. It contains a peramivir hydrate.

Peramivir Use and Manufacturing

A new antiviral agent for influenza treatment; it can be used as neuraminidase inhibitor for treating human and avian influenza

Drug Function and Efficacy

Peramivir is a cyclopentane anti-influenza virus drug that can bind to the active site of influenza virus neuraminidase and has inhibitory activity against human influenza A and B viruses. Biochemical analysis showed that peramivir could inhibit the neuraminidase activity of several influenza A and B virus strains, with a median ICso value of 0.2nM (0.09~1.4nM, n=15) for influenza A virus strains, a median ICco value of 1.3nM (0.06~11nM, n=8) for influenza B virus strains, and an ICso value range of 0.06~0.26nM for 2009 HINIA influenza virus strains (swine flu). The antiviral activity of peramivir against laboratory virus strains and clinically isolated virus strains was investigated in cell culture experiments. The ECs0 value for seasonal influenza A HIN1 strain is 1μM (0.09~21μM, n=5), the ECso value for influenza A H3N2 strain is 0.07μM (0.01~0.16μM, n=12), and the ECso value for influenza B strain is 2.2μM (0.06~3.2uM, n=5). Limited biochemical analysis, cell culture and animal model data show that the antiviral activity of peramivir and oseltamivir is synergistic.
