Description

Labetalol hydrochloride is a mixed alpha/beta adrenergic antagonist that is used to treat high blood pressure.


Labetalol hydrochloride is a member of salicylamides.|Labetalol Hydrochloride is the hydrochloride form of labetalol, a third generation selective alpha-1-adrenergic antagonist and non-selective beta-adrenergic antagonist with vasodilatory and antihypertensive properties. Labetalol competitively binds to alpha-1-adrenergic receptors in vascular smooth muscle, thereby inhibiting the adrenergic stimulation of endothelial cell function and vasoconstriction in peripheral blood vessels. This agent also binds to beta-receptors in the bronchial and vascular smooth muscle, resulting in a decrease in adrenergic stimulation. The result is a decrease in resting and exercise heart rates, cardiac output, and in both systolic and diastolic blood pressure, thereby resulting in vasodilation, and negative chronotropic and inotropic cardiac effects.|A salicylamide derivative that is a non-cardioselective blocker of BETA-ADRENERGIC RECEPTORS and ALPHA-1 ADRENERGIC RECEPTORS.

Drug Function and Efficacy

It has both alpha and beta receptor blocking effects, with weak intrinsic activity and membrane stabilizing effects. The effect of blocking beta receptors is 4-8 times that of blocking alpha receptors; the effect of blocking beta1 receptors is 1/4 of propranolol, and the effect of blocking beta2 receptors is 1/17-1/11 of propranolol. At equivalent doses, its heart rate slowing effect is milder than that of propranolol, and its antihypertensive effect occurs faster. The vasodilation caused by blocking alpha receptors is also involved in the antihypertensive and anti-anginal mechanisms. Compared with simple beta receptor blockers, it has a stronger antihypertensive effect during standing and exercise tests. In addition, it can increase renal blood flow, while propranolol reduces it.
