Methods of Manufacturing

The industrial production of lysine mainly uses the direct fermentation method of microorganisms. Lysine is an amino acid in the branch of aspartate, and aspartate kinase is a key enzyme. It is inhibited by the coordinated feedback of lysine and threonine. A single amino acid cannot completely inhibit enzyme activity. Almost all strains used are Corynebacterium glutamicum, which is a homoserine auxotrophic mutant strain and lacks homoserine dehydrogenase. Since aspartate kinase is not inhibited by a single amino acid, homoserine-deficient mutant strains limit the production of threonine and relieve its feedback inhibition. Therefore, lysine fermentation is carried out, and the sugar yield of the production strain can reach more than 40%. According to the Chinese Academy of Sciences Microbiology Research, the homoserine-deficient strain A.S.1.563 has a pilot-scale acid production rate of 3.66%, a sugar yield of 30%, and sugar cane molasses up to 42%. The strain was activated at 30-32°C for 24 hours, cultivated on a slope under the same conditions, and then cultured with primary seeds. The seed medium is: glucose 2%, corn steep liquor 2%, magnesium sulfate 0.05%, ammonium sulfate 0.4%, dipotassium hydrogen phosphate 0.1%, soybean cake hydrolysate 1%, inoculum volume 5%, culture at 32°C for 17 hours. Fermentation The composition of the fermentation medium is: glucose 15%, ammonium sulfate 2%, dipotassium hydrogen phosphate 0.1%, magnesium sulfate 0.04%, soybean cake hydrolysate 2%, and urea 0.4%. The inoculum was 5%, cultured at 32°C for 38 hours, and the required aeration rate was 1:0.3 V/(V·min), stirring, and paying attention to the biochemical metabolism curve during the fermentation process. Adsorption, elution, concentration, and crystallization. Heat the fermentation broth at 80℃ and stir it fully for 10 minutes. When it is cooled to 40℃, take samples to measure the acid content, and add H2SO4 to adjust the pH to 4-5. After precipitation for 2 hours, introduce into 732 -NH4+ resin column. When the acid content of the fermentation broth is about 2.5%, the ratio of resin volume to fermentation broth volume is 1:3. The flow rate of the upper column is controlled at 1000 ml/min. When the pH value of the effluent gradually drops to 6-5.5 and the acid content is 0.5%, the resin is saturated. Generally adsorb 2-3 times. The saturated resin is washed with no salt water to remove the bacteria and impurities until the effluent is clear. Then use 2.5 mol/L ammonia to hydrolyze and absorb at a flow rate of 700-800 ml/min. Start collecting at pH 8, which is about 1.5-2 times the volume of the resin. Stop collecting when the eluent pH is 13-14. The obtained eluate is heated to remove ammonia, the thin film is vacuum concentrated to 21°Be, cooled, concentrated HCl is added to adjust the pH to 4.9, the crystal is allowed to stand for 3 days, the crystal is filtered, dried to obtain a crude product, the mother liquor is concentrated and recovered. Recrystallization and refining The crude product is dissolved in distilled water and prepared to 12°Be, and then 10%-12% by weight of the crude product is added with activated carbon (it is required that the activated carbon does not contain Ca2+, Mg2+, for this purpose, use dilute HCl to pre-treat the activated carbon). The filtrate is filtered, concentrated under reduced pressure (controlled at about 93.33kPa), and the humidity is 40-45°C until it is saturated. Allow it to cool naturally to crystallize, filter the crystals, and dry at 60°C to obtain crude L-lysine salt with a yield of 50%.

Uses
