Methods of Manufacturing

General procedure: To a solution of carbamoyl chloride (0.452 mmol) in DCM (5 mL) was successively addedtriethylamine (126 mL, 0.908 mmol, 2 eq.), 4-DMAP (8 mg, 0.067 mmol, 15%) and amine(0.673 mmol, 1.5 eq.). The reaction mixture was stirred at rt for 1h to 12h. It was then washedwith saturated aqueous NaHCO3 solution, water, dried over MgSO4 and concentrated underreduced pressure to give crude urea that was further purified by flash chromatography orpreparative TLC.General procedure: To a solution of azetidine (5.81 mmol) in DCM (20 mL) cooled to 0C was added portionwiseBTC (661 mg, 2.32 mmol, 1/3 eq.x1.2). The reaction mixture was stirred at 0C for 0.5h andallowed to reach rt. After 0.5h to 24h, the reaction mixture was then concentrated underreduced pressure and the crude residue was purified by flash chromatography.
