Description

Thymopentin is a biologically active peptide secreted mainly by the epithelial cells of thymic cortex and medulla. Thymopentin is an effective immunomodulatory agent with a short plasma half-life of 30 seconds. Thymopentin enhances the generation of T-cell lineage derived from human embryonic stem cells (hESCs)[1][2].

Thymopentin is an oligopeptide.|Thymopentin is a synthetic pentapeptide which is the active site of the naturally occurring hormone thymopoietin with immunomodulating properties. Thymopentin enhances the production of thymic T cells and may help restore immunocompetence in immunosuppressed subjects. This agent also augments the effects of ionizing radiation by arresting cancer cells in the G2/M phase of the cell cycle. (NCI04)|Synthetic pentapeptide corresponding to the amino acids 32-36 of thymopoietin and exhibiting the full biological activity of the natural hormone. It is an immunomodulator which has been studied for possible use in the treatment of rheumatoid arthritis, AIDS, and other primary immunodeficiencies.

Drug Information

Substances that augment, stimulate, activate, potentiate, or modulate the immune response at either the cellular or humoral level. The classical agents (Freund's adjuvant, BCG, Corynebacterium parvum, et al.) contain bacterial antigens. Some are endogenous (e.g., histamine, interferon, transfer factor, tuftsin, interleukin-1). Their mode of action is either non-specific, resulting in increased immune responsiveness to a wide variety of antigens, or antigen-specific, i.e., affecting a restricted type of immune response to a narrow group of antigens. The therapeutic efficacy of many biological response modifiers is related to their antigen-specific immunoadjuvanticity. (See all compounds classified as Adjuvants, Immunologic.)

Thymopentin Use and Manufacturing

Methods of Manufacturing

Preparation of thymosin-II. Raw material treatment and concentration The calf thymus was used as raw material, 0.1mol/L NH4HCO3 was added, and the mixture was extracted to obtain alkaline protein solution, which was heated to 70°C to remove denatured contaminated protein, remove the precipitate, and concentrate the supernatant. Calf thymus [0.1 mol/L NH4HCO3]→Basic protein solution [70℃, deproteinized protein]→[Concentrate] Concentrate is separated, chromatographed, purified by SephadexG-50 and hydroxyphosphorus lime column chromatography, separated, then After QAE-Sephadex column chromatography, it is purified to obtain thymosin-II concentrated solution [Sephadex G-50]→[hydroxyphosphorus lime column] chromatographic solution [QAE-Sephadex column]→thymosin-II thymosin -II extraction raw material processing, ultrafiltration, centrifugation using fresh bovine spleen as raw material, adding 0.1 mol/L NH4HCO3, homogenous extraction, to obtain alkaline soluble protein. Ultrafiltration with H10×100, the filtrate was concentrated, the water bath was heated to 70 ℃, centrifuged for 15 min, leaving the supernatant. Fresh bovine spleen [0.1 mol/L NH4HCO3] → alkaline protein solution [ultrafiltration, concentration] → [70°C, centrifugation for 15 min] The supernatant was purified through the column, the supernatant was adjusted to pH 5.2, and cation exchange resin (50 mg/ml), stir and filter, then adjust the filtrate to pH 9.5, add cation exchange resin (50mg/m1), stir and filter to obtain filtrate. Supernatant [pH5.2, cation column] → filtrate [pH9.5, cation column] → filtrate gel filtration, purification The filtrate is filtered through Sephadex G-75 gel to obtain active TP and pantothenic hormone, and then anion exchange Resin chromatography (NH4HCO3, ionic strength change 0.1-0.8mol/L), to get TP-III, heteroproteins and pan hormones. Filtrate [Sephadex G-75 gel filtration] → active TP and ubiquitin [anion resin chromatography] → TP-III, heteroprotein and ubiquitin.
