Fmoc-L-Dapa-OH Use and Manufacturing

Methods of Manufacturing

To a stirred solution of sodium bicarbonate (0.154 g)and Na-Fmoc, L-diamino propionic acid (0.5 g, 1.53 mmol) in 1: 1 water and acetonitrile mixture (4 mE) was added 4-chloro-7-nitrobenzo-2-oxa-l , 3-diazole (0.367 g, 1.836 mmol). The reaction mixture was allowed to stir overnight. Solvent was removed using rotary evaporation, and the remaining crude was purified by flash chromatograph on silica gel (CH2C12: MeOH=l0:3) with trance acetic acid to give product as brown solid (0.482 g, 0.985 mmol, 64.3%). 'H NMR (400 MHz, DMSO, 25 C.) oe 3.80 (bs, 2H), 4.15 (t, J=8.4 Hz, 1H), 4.28 (d, J=9.2 Hz, 2H), 4.37 (m, 1H), 6.48 (d, J=ll.6 Hz, 1H), 7.24 (t, J=9.6 Hz, 2H), 7.35 (m, 2H), 7.60 (d, J=l0.0 Hz, 2H), 7.63 (d, J=l0.4 Hz, 1H), 7.85 (d, J=l0.0 Hz, 2H), 8.48 (d, J=l0.4 Hz, 1H).
