1-(2-Chloroethyl)pyrrolidine hydrochloride Use and Manufacturing

Methods of Manufacturing

165 ml (2 mol) of tetrahydropyrrole, 67 ml (1 mol) of 2-chlorohydrin and 200 ml toluene were added successively into a reaction flask. The mixture was heated to the reflux temperature and reacted for 3 h. After cooling to room temperature, a solid was separated out. The solid was filtered by sucking, and the filter cake was washed with 20 ml toluene. The temperature of the filtrate was controlled at about 75° C., and 200 ml thionyl chloride was added dropwise. After refluxing for 2 h, the mixture was cooled to room temperature and then concentrated to dry in a reduce pressure. The residue was recrystallized with absolute ethyl alcohol to obtain 105 g white solid with the yield of 62.3percent. m.p. 198-203° CThe 4.6 ml pyrrolidine (57.5 mmol) with 4.6 ml 2 - chloro ethanol (69.0 mmol) is added to the 40 ml in toluene, heating to reflux 2 hours, the reaction is cooled down to the room temperature, under ice bath instillment chlorination sulfoxide 8.4 ml (114 mmol), 20 min drop end, removed ice, stirring at room temperature for 6 hours, evaporating the toluene and thionyl chloride under reduced pressure, the residual solid using ethanol to recrystallize the, results in the chlorine ethyl pyrrolidine hydrochloride 8.3 g.2- pyrrolidin-1-ylethanol (6.24 g, 0.05 mol) was stirred while cautiously adding thionyl chloride (40 ml_, 0.54 mol). The stirred mixture was refluxed for 1 hour under nitrogen atmosphere, then concentrated in vacuo. The residue was triturated with diethyl ether to give a solid, and the liquid decanted; this process was repeated several times, until the decantate was almost colorless. The flask containing solid was evaporated to give a free- flowing, cocoa-colored solid (4.21 g, 46percent).

Uses

1-(2-Chloroethyl)pyrrolidine is a haloalkyl substituted pyrrolidine used in the preparation of various pharmaceutical compound such as the estrogen receptor antagonist Nafoxidine (N212800) and its ana logues.
