Benzo[b]thiophene, 7-fluoro- (9CI) Use and Manufacturing

To a solution of 4-5 (4 g, 20.4 mmol) in quinoline (21 mL) was added Cu powder (440 mg, 6.12 mmol) at rt. The mixture was heated to 185C and stirred for 2 h. The solution was cooled to rt, poured into 2N HC1 solution and extracted with DCM. The organic phase was concentrated (< 40C) to give crude 4-6 (3.3g, 74.2%).General procedure: To a solution of 2-chlorobenzenethiol (1.53 g, 10.58 mmol, 1.0 eq.) and 2-bromo-1, 1-diethoxyethane (2.4 mL, 15.87 mmol, 1.5 eq.) in acetone (10 ml) was added potassium carbonate (3.65 g, 26.45 mmol, 2.5 eq). The mixture was refluxed for 90 minutes and then 0, 5 eq of 2-bromo-1, 1-diethoxyethane was added. The mixture was refluxed for another 2 hours, cooled to room temperature and concentrated. The resulting residue was purified by flash column chromatography on silica gel using hexane/ diethyl ether (2%) as eluent to afford (2-chlorophenyl)(2, 2-diethoxyethyl)sulfane as a light yellow oil (2.71 g, 97%). (0142) To a solution of polyphosphoric acid (6.0 g, 51.97 mmol, 9 eq.) in toluene (15 ml) was added under vigorous stirring a solution of (2-chlorophenyl)(2, 2-diethoxyethyl)sulfane (1.5 g, 5.77 mmol, 1 eq) in toluene (5 mL). The mixture was stirred at reflux temperature for 5 hours and was then poured into ice cold water (30 mL) and stirred for 10 minutes. The mixture was washed with toluene, sat. aq. bicarbonate solution and brine, dried (Na2SO4) and concentrated. The resulting residue was purified by flash column chromatography on silica gel using hexane as eluent (as described in WO 2009/008992 A2) to afford 7-chlorobenzo[b]thiophene as a light-yellow oil (0.68 g, 70%). 1H-NMR (300 MHz, CDCl3-d) delta (ppm): 7.77 - 7.71 (m, 1H), 7.50 (d, J = 5.4 Hz, 1H), 7.41 - 7.30 (m, 3H).[0656j To a mixture of aluminum trichloride (2.66 g, 20 mmol) in dry DCM (10 mL) cooled at 0C under N2, was added a solution of To a solution of Step 1 3-[(7-Fluoro-benzo[b]thiophen-2-yl)-hydroxy-methyl]-3-(tetrahydro-pyran-4-ylmethyl)-pyrrolidine-1-carboxylic acid tert-butyl ester To a solution of Chlorobenzene (25 mL) was heated to reflux and polyphosphoric acid (1 mL) was added. The 1-(2, 2-dimethoxy-ethylsulfanyl)-2-fluoro-benzene was then added slowly turning the solution dark. After 3 hours of heating, the reaction was cooled to room temperature and diluted with water (50 mL). The solution was extracted with benzene (2*50 mL). The extracts were concentrated and chromatographed (0 to 15% ethyl acetate in hexane) to yield 2-Fluorothiophenol (4.14 g, 32.6 mmol) was dissolved in anhydrous THF (100 mL). Potassium tert-butoxide (1.0 M in THF, 35.8 mL) was added and the suspension was stirred at room temperature for 15 minutes. 2-Chloroacetaldehyde dimethyl acetal was added and the mixture was stirred for 3 days. Water (100 mL) was added and the solution was extracted with diethyl ether (3×100 mL). The extracts were concentrated to a yellow oil and chromatographed (5 to 20% ethyl acetate in hexane) to yield 1-(2, 2-dimethoxy-ethylsulfanyl)-2-fluoro-benzene (6.42 g) as a colorless oil. Chlorobenzene (25 mL) was heated to reflux and polyphosphoric acid (1 mL) was added. The 1-(2, 2-dimethoxy-ethylsulfanyl)-2-fluoro-benzene was then added slowly turning the solution dark. The extracts were concentrated and chromatographed (0 to 15% ethyl acetate in hexane) to yield 7-fluorobenzothiophenyl (0.77 g). The 7-fluorobenzothiophenyl (0.77 g, 5.1 mmol) and dichloromethyl methyl ether (0.872 g, 7.6 mmol) were dissolved in anhydrous DCM (25 mL). Titanium tetrachloride (1.0 M in DCM, 7.6 mL, 7.6 mmol) was added, turning the solution dark. After 30 minutes at room temperature, the reaction was poured into a mixture of saturated aqueous NaHCO3 and ice. The mixture was stirred for about 30 minutes and then was extracted with DCM (2×50 mL).
