Preparation of N-\C\ S)-2-((1 -f(2-Cvanophenyl)sulfonyllhexahydro-1 /-/-azepin-4-yljamino)- i -fcvclopentylmethvD^-oxoethyll-i -benzothiophene^-carboxamide; a) 1 , 1 -Dimethylethyl 4-({[(phenylmethyl)oxy]carbonyl}amino)hexahydro-1 H-azepine- 1 -carboxylate; To a CH2CI2 (50 ml_) solution of 1 , 1-dimethylethyl 4-aminohexahydro-1 H-azepine- 1 -carboxylate 5 (2.85 g, 13.32 mmol) was added benzylchloroformate (2.30ml, 15.98 mmol) and TEA (5.3ml, 40.0 mmol), and the mixture was stirred at room temperature for 4 hours. The solution was washed with saturated NaHCO3 and brine, dried (MgSO4), filtered and concentrated to a solid. Purification by column chromatography (30percent-70percent EtOAc/hexanes) gave the title compound as a white solid in 82percent yield (3.8 g): LCMS (M+H): 349.Example 83
