6-BROMO-2,4-DICHLOROQUINAZOLINE Use and Manufacturing
A solution of 25 (12.0g, 49.8mmol), POCl3 (46.4 mL, 498 mmol) and N, N-diethylaniline (31.7 mL, 199 mmol) was heated under reflux for 5h. After cooling, the mixture was evaporated and the mixture was diluted with H2O and CHCl3. The organic layer was washed with H2O and sat. NaCl, then dried over MgSO4 and filtered. After removal of the solvent in vacuo, the residue was purified by silica gel column chromatography (CHCl3/hexane; 1:2 to 4:1) to give the title compound as white solid (10.4 g, 75percent). 1H NMR (CDCl3) δ = 7.88 (dd, 1H, J = 0.3, 9.0 Hz), 8.06 (dd, 1H, J = 2.1, 9.0 Hz), 8.43 (d, 1H, J = 0.3, 2.1 Hz).
