3-BROMO-5-METHYLBENZONITRILE Use and Manufacturing

3-Bromo-5-methylbenzonitrilePhosphorous pentoxide (29.8 g, 210.2 mmol) was added to a suspension of 3-bromo-5- methylbenzamide (15.0 g, 70.1 mmol) in CHClIntermediate 503-Bromo-5-methylbenzonitrile; A mixture of 1, 3-dibromo-5-methylbenzene (1.0 g, 4.0 mmol), copper cyanide (0.179 g, 2.00 mmol), pyridine (0.323 mL, 4.00 mmol), and DMF (15 mL) were heated at 190° C. for 10 h in microwave reactor. The reaction mixture was allowed to cool to r.t., and then poured into a solution of HTo a sealable vessel was added 2-(3-bromo-5- methylphenyl)-4, 4, 5, 5-tetramethyl-l, 3, 2-dioxaborolane (26.1 g, 88 mmol), copper(II)nitrate trihydrate (42.5 g, 176 mmol), zinc cyanide (31.0 g, 264 mmol), cesium fluoride (13.35 g, 88 mmol), 125 mL methanol, and 50 mL water. The vessel was sealed and heated to 100 °C for 5 hours. The reaction mixture was cooled and an insoluble tan solid was filtered and washed with ethyl acetate and water. The layers were separated and the aqueous layer was extracted with ethyl acetate. The organic layers were washed with brine, dried over magnesium sulfate, filtered and evaporated. The residue was purified by column chromatography eluting with 5 and 10 percent ethyl acetate/hexane (3.4 g, 20percent)
