Methods of Manufacturing

Condensation with meta-chloroaniline and triethyl orthoformate to produce bis(m-chlorophenyl) formamidine, and then condenses with diethyl malonate and triethyl orthoformate under the catalysis of boron trifluoride to obtain meta-chloroaniline Diethyl methylenemalonate is then cyclized in paraffin oil at high temperature of 270-280°C to synthesize 7-chloro-4-hydroxyquinoline-3-carboxylic acid ethyl ester. The latter is obtained by alkali hydrolysis and neutralization.

Uses

This product is heated to decarboxylate to obtain 4-hydroxy-7-chloroquinoline, which is used to synthesize chloroquine phosphate.
