4-CHLORO-3-IODO (1H)INDAZOLE Use and Manufacturing

Preparation of 4-chloro-3-iodo-lH-indazole[0440] To a solution of 4-chloro-lH-indazole (1.7 g, 11.2 mmol, 1.0 eq) in DMF (20 mL), was added KOH (1.25 g, 22.4 mmol, 2.0eq). The mixture was stirred at room temperature for 30 minutes. To the resulting mixture, was added IIodine (21 g, 82.74 mmol, 2.00 equiv) was added dropwise into a solution of 4-chloro-1H-indazole(6.3 g, 41.29 mmol, 1.00 equiv) and potassium hydroxide (8.4 g, 149.72 mmol, 3.60 equiv) inN, N-dimethylformamide (100 mL). The resulting solution was stuffed overnight at roomtemperature, quenched with 200 mL of sat. aq. Na2S2O3, extracted with ethyl acetate, dried overanhydrous sodium sulfate, and concentrated under vacuum. The residue was purified by a silica gel column with ethyl acetate/petroleum ether (1:5) to give 5 g (43percent) of the title compound as a yellow solid. LC-MS (ES, m/z): 279[M+H].
