Drug Function and Efficacy

Inhibitory effect on epidermal cell proliferation; Tacalcitol inhibits DNA synthesis and cell proliferation in rat epidermal culture cells and normal or psoriasis-derived human epidermal culture cells. Tacalcitol inhibits the activity of DOC (ornithine decarboxyl alcohol) in the epidermis of hairless rats that stimulate cell proliferation by applying TPA (12-O-tetradecanoylphospholipid). And by applying the ointment to psoriasis patients for 4 weeks, it can inhibit DNA synthesis and cell division, reduce S-phase cells, and inhibit epidermal cell proliferation. Differentiation-inducing effect on epidermal cells: For rat epidermal culture cells, Tacalcitol promotes the formation of intracellular insoluble membrane (cornified envelope) and increases transglutaminase (Tgase) activity. For the epidermis of hairless rats, Tgase activity is increased. For normal human cultured epidermal cells, Tacalcitol promotes the synthesis of invogulin, a precursor protein shell of intracellular insoluble membrane. In addition, the skin of the lesion site of psoriasis patients after application was observed under an electron microscope, and normal keratinization tendency such as the formation of a protein pattern (keratin pattern) of the stratum corneum and the formation of a granular layer with epidermal transparent keratin (keratohyalin) was found. It has affinity for the specific protein receptor of 1α, 25-(OH)2D3 of epidermal cells.
