to a mixture of [0840] to a solution of NAH (1.46 g, 36.59 mmol, 60% purity) in THF (80 ml) was added A solution of Potassium carbonate (890 mg) was added to an N, N-dimethylsulfoxide solution (3 ml) of methyl 4-bromo-1 H-pyrazole-3-carboxylate (CAS No. 81190-89-8) (600 mg), followed by stirring at room temperature for 10 minutes did.2, 2, 2-Trifluoromethyl trifluoromethanesulfonate (0.84 ml) was added to the reaction solution, and the mixture was further stirred for 18 hours.The reaction solution was purified by column chromatography to give the title compound (230 mg).General procedure: Aryl Halide, tetrakis (triphenylphosphine)palladium or Palladium (II)bis(triphenylphosphine) dichloride (0.05eq) and boronic acid or pinnacol ester (1.2eq) were weighed out into a microwave vessel or sealed tube. Acetonitrile (3mL/mmol) and a 1M aqueous solution of Sodium Carbonate (3eq) were added. The vessel was capped and heated thermally 3-18hrs at 100C. Upon completion, the reaction was cooled and crude product was either triterated via addition of water and collection by filtration or extracted with sat ammonium chloride and DCM. If the crude product was an intermediate, it was taken into the next step in most cases w/o further purification or alternatively submitted for reverse phase HPLC purification when it was a final product.Similar to the procedure as described in General Procedure M, methyl 4-bromo-lH- pyrazole-3-carboxylate was reacted with l, 3-dimethyl-4-(4, 4, 5, 5-tetramethyl-l, 3, 2- dioxaborolan-2-yl)-lH-pyrazole to give the title compound (200 mg, 20%) as yellow oil. LC-MS (ES, m/z): 221 [M+H]+.
