Lysozyme Use and Manufacturing
Raw material processing: Take 70kg of fresh or frozen egg white to make the pH around 8. Thaw and sieve to remove the umbilical cord, eggshell fragments and other impurities in the egg white. Lower the temperature to about 50 (not more than 100), add 724 resin 10kg (wet weight) while stirring, wait until the resin is all suspended in the egg white, keep it at 0-50, stir and absorb for 5h, and then move the egg white into the freezer for 0-50 , Let stand overnight. Egg white 724 resin; 30C→adsorbing resin to remove impurities, elution, and sediment. Pour out the supernatant, wash off the protein attached to the resin surface with water, and wash it slowly and repeatedly 4 times to prevent the loss of the fingers. The resin is suction filtered to remove water. Add 24L, pH6.5 phosphoric acid (0.15mol/L) buffer solution to the resin in 3 times. After each addition, stir for about 15min. After stirring, filter under reduced pressure to remove water. Then 18L of ammonium sulfate (100g/L or 10%) was divided into 4 times to elute the lysozyme. After each addition, it was stirred for 0.5h, filtered and drained. Combine the eluents and add 320g solid ammonium sulfate to each liter of eluent to make the content reach 400g/L (40%). At this time, a white precipitate will appear. Let it stand overnight in a cold place. The supernatant will be siphoned the next day to precipitate. Centrifugal separation to obtain crude product. Resin adsorbent H2O; Phosphate buffer → Detergent protein adsorbent (NH4)^2^) SO4 → Eluent (NH4) 2SO4 → Precipitation crude product dialysis Add the crude product to 1.5 kg of distilled water to completely dissolve it, and put it into the dialysis bag Dialysis in the cold storage for 24 to 36 hours to remove most of the ammonium sulfate to obtain dialysate. Crude distilled water; dissolve→dialysis dialysate salting out to clarify the dialysate, slowly add sodium hydroxide (1mol/L) dropwise under constant stirring to neutralize excess acid, and make the pH reach 8.5-9, if there is precipitation , Should be removed by centrifugation immediately, and then add hydrochloric acid (3mol/L) under constant stirring to make the pH of the solution reach 3.5. Slowly add 5g of solid sodium chloride per 100ml of acidic dialysate by volume, that is, a white precipitate will be precipitated. Place 50 cold storage for 2 days, centrifuge or filter to get lysozyme precipitation. Dialysate^...→NaCl salting out wet lysozyme drying while stirring continuously, add 10 times the amount of anhydrous acetone cooled to 00C according to the mass of the lysozyme precipitation, wait until the particles are fine, and place it in a cold place for several hours. Remove the acetone and dry the precipitation in a vacuum until there is no odor of acetone to obtain the raw material for oral or topical lysozyme. Wet lysozyme acetone; 00C→Dry lysozyme preparation Take dry crushed sugar powder, add 50g of talc powder per kilogram of powdered sugar (that is, 5%), pass through a 129-mesh sieve, and add starch slurry (50g/L or 5%) ) Appropriate amount, stir evenly in the mixer, make soft material, pass through a 12-mesh sieve, dry at 700C, use a 14-mesh sieve to whole, and control the water content at about 2% to 4%. Add a certain amount of lysozyme powder to mix, add 1% stearic acid enzyme, pass through a 16-mesh sieve twice, and press tablets to obtain lysozyme buccal tablets, each containing 20mg lysozyme.
