Methods of Manufacturing

Phosphorus pentasulphide (1.16 g, 5.2 mmol) in tetrahydrofuran was added to a stirred solution of 3-hydroxy-4H-pyran-4-one (Compound II) (14.2 kg) and methanol (70 L)Add to the reaction kettle and stir. Cool down to 5 C, add sodium hydroxide (5.6 Kg) and water(18 L) of a mixed solution. After stirring for 1 h, add 37%Formaldehyde (25L).Warming up to room temperature24h, the reaction was completed, the temperature was lowered to 5 C, and the pH was adjusted with concentrated hydrochloric acid.The methanol was removed by concentration under reduced pressure, and dichloromethane (EtOAc) was evaporated. Then filter the inorganic salts, Liquid separation. The aqueous phase was extracted twice more with dichloromethane (45 L).The combined organic phases are concentrated under reduced pressure, and the solid is evaporated by isopropanol, and filtered under cooling.get3-hydroxy-2-(hydroxymethyl)-4H-pyran-4-one (Compound III)15.5Kg, purity 96%, yield 86%
