Description

ChEBI: The trisodium salt of phosphonoformic acid. It is used as an antiviral agent in the treatment of cytomegalovirus retinitis (CMV retinitis, an inflamation of the retina that can lead to blindness) and as an alternative to ganciclovir for AIDS patients who r quire concurrent antiretroviral therapy but are unable to tolerate ganciclovir due to haematological toxicity.


Trisodium phosphonoformate is the trisodium salt of phosphonoformic acid. It is used as an antiviral agent in the treatment of cytomegalovirus retinitis (CMV retinitis, an inflamation of the retina that can lead to blindness) and as an alternative to ganciclovir for AIDS patients who require concurrent antiretroviral therapy but are unable to tolerate ganciclovir due to haematological toxicity. It has a role as an antiviral drug. It is an organic sodium salt and a one-carbon compound. It contains a phosphonatoformate.|An antiviral agent used in the treatment of cytomegalovirus retinitis. Foscarnet also shows activity against human herpesviruses and HIV.

Methods of Manufacturing

Method 1: In the absence of organic base, phosphorus trichloride and ethanol produce diethyl phosphite; in the presence of organic base, triethyl phosphite is obtained. After the reaction between triethyl phosphite and ethyl chloroformate at 160℃, the fractions of 148~150℃/2.40kPa are collected to obtain triethylphosphonate with a yield of 71%. And diethyl phosphite in anhydrous ether, in the presence of sodium metal, and ethyl chloroformate reaction at room temperature, can also get triethyl phosphonate, yield 80%. 21 parts of triethyl phosphonate were dissolved in 60 parts of water containing 24 parts of 50% sodium hydroxide and refluxed for 2 hours. After cooling overnight, 4.7 parts of sodium phosphonate hexahydrate were filtered, melting point> 250°C. Method 2: Newly prepared sodium cyanide was placed in carbon tetrachloride, and equimolar diethyl phosphite was added dropwise at about 10°C, and then reacted at 35°C for 3 hours. Filtration, concentration and distillation under reduced pressure to obtain diethyl cyanophosphate, yield 72%, boiling point 88~91℃/1.46kPa. It was refluxed with 40% sodium hydroxide for 2h, filtered, and recrystallized with water to obtain sodium foscarnet hexahydrate with a yield of 52% and a melting point of >250°C.
