Revolutionary Breakthrough: Hengrui Pharmaceuticals Unveils Groundbreaking Findings on Carrelizumab for Late-Stage Lung Cancer
In a significant development for late-stage lung cancer treatment, Hengrui Pharmaceuticals has recently unveiled the latest data from their innovative drug, Carrelizumab. The findings, presented at the 2024 European Lung Cancer Conference, shed light on the long-term efficacy of Carrelizumab in combination with carboplatin and pemetrexed as a first-line therapy for advanced driver gene-negative non-squamous non-small cell lung cancer (NSCLC).
Hengrui Pharmaceuticals' groundbreaking study, known as the CameL trial, demonstrated remarkable outcomes in patients with late-stage NSCLC. The five-year overall survival (OS) rate for patients receiving Carrelizumab in combination with chemotherapy reached an impressive 31.2%, compared to 19.3% in the chemotherapy-only group. These findings hold great promise in extending the lives of patients, with a substantial 45% reduction in the risk of disease progression observed through the four-year and five-year progression-free survival (PFS) rates, which stood at 16.1%.
The five-year survival rate is a crucial indicator for assessing the clinical cure of cancer. The "Healthy China Action - Cancer Prevention and Control Program" has identified improving the five-year survival rate as a primary goal for cancer treatment. Notably, the CameL trial revealed that nearly one-third of advanced lung cancer patients in the Carrelizumab combination therapy group achieved a survival period exceeding five years. This achievement underscores Hengrui Pharmaceuticals' commitment as a leading Chinese innovator in the field of oncology and their dedication to prolonging the lives of cancer patients.
According to the latest data released by the National Cancer Center, lung cancer remains the leading cause of malignancies and mortality in both men and women. In 2022, there were approximately 1.06 million new cases of lung cancer, accounting for 22.0% of all malignant tumors, and resulting in 733,000 deaths, representing 28.5% of total cancer-related deaths. Lung cancer imposes a significant burden on patients and their families, both in terms of health and economic consequences. Unfortunately, most lung cancer cases are diagnosed at an advanced stage, with less than 10% of patients surviving beyond five years under traditional chemotherapy treatments.
Carrelizumab, a humanized IgG4 monoclonal antibody targeting PD-1, has demonstrated remarkable efficacy in the treatment of various solid tumors, including lung cancer, liver cancer, esophageal cancer, and nasopharyngeal carcinoma, by significantly prolonging overall survival.
The CameL trial, led by Professor Zhou Caicun from the Oriental Hospital affiliated with Tongji University, is a randomized, open-label, multicenter, Phase III clinical trial. In this trial (NCT03134872), Carrelizumab in combination with carboplatin and pemetrexed was compared to carboplatin and pemetrexed alone as a first-line treatment for previously untreated advanced non-squamous NSCLC patients without EGFR/ALK mutations. The latest survival results, reported at the 2024 European Lung Cancer Conference (ELCC) after approximately five years of follow-up, revealed that the median overall survival was 27.1 months in the Carrelizumab combination therapy group, compared to 19.8 months in the chemotherapy-only group (HR 0.74 [95% CI, 0.58-0.93]; P=0.0043). The Carrelizumab combination therapy group also demonstrated a higher five-year OS rate of 31.2% compared to 19.3% in the chemotherapy group.
Moreover, 52.7% of patients in the chemotherapy group crossed over to receive immunotherapy after disease progression, with 45.9% of these patients receiving Carrelizumab. After adjusting for the impact of crossover treatments, the Carrelizumab combination therapy group exhibited even more significant OS benefits (adjusted HR 0.62 [95% CI, 0.49-0.79]; P<0.0001). Consistent with previous reports, the Carrelizumab combination therapy also demonstrated improved progression-free survival (PFS) compared to chemotherapy alone (median PFS: 11.0 months vs. 6.5 months; HR 0.55 [95% CI, 0.44-0.68]; P<0.0001).
The latest data released by Hengrui Pharmaceuticals on their Carrelizumab therapy for late-stage lung cancer signifies a breakthrough in the field of oncology. With its remarkable long-term survival rates and effective disease control, Carrelizumab in combination with chemotherapy offers new hope for patients with advanced non-squamous NSCLC.
