Propylthiouracil is a thiourea antithyroid drug that mainly inhibits the synthesis of thyroxine. Its mechanism of action is to inhibit the oxidation of iodide taken into the thyroid gland and the coupling of tyrosine by inhibiting thyroid endoperoxidase, thereby hindering the synthesis of thyroid hormone (T4) and triiodothyronine. This product does not block the release of stored thyroid hormone, so it only has a significant clinical effect after the thyroxine in the body is eliminated and the concentration in the circulation decreases. In addition, propylthiouracil also inhibits the deiodination of T4 in peripheral tissues to produce T3. This product is a white compressed tablet with a bitter taste.

The usual dosage is 1-2 tablets at a time, 3-6 tablets a day. The maximum dosage is 4 tablets at a time, 12 tablets a day. Medical treatment of hyperthyroidism: The first dose for adults is 6-12 tablets/day, divided into 3-4 times, and then gradually reduced to a maintenance dose of 0.5-2 tablets/day after the symptoms are controlled. The course of treatment is about one and a half years. Preparation before thyroid surgery: To reduce anesthesia and postoperative complications and prevent the induction of thyroid crisis, this product should be used before surgery. Thyroid crisis: 16-20 tablets/day, divided into 3-4 times, for 7 consecutive days, and then switch to the usual dose after the crisis is controlled.

Common reactions include joint pain, fever, erythema, skin pigmentation, itching, hair loss, loss of appetite, nausea and vomiting, diarrhea, headache, and lethargy. It can cause leukopenia and agranulocytosis, rash and other allergic reactions. Some people have a tendency to exfoliative dermatitis, so the drug should be discontinued immediately.

It is contraindicated for patients with severe liver damage, severe leukocyte deficiency, and allergy to thiourea drugs.

Precautions for children: Hypothyroidism should be avoided during medication for children. Precautions for pregnancy and lactation: Use with caution for pregnant women and prohibit use for lactating women. Precautions for the elderly: The dosage should be reduced for the elderly, especially those with renal impairment. If hypothyroidism is found, thyroid tablets should be added.

Combination use of coumarin anticoagulants may lead to bleeding tendency.
