Function and Efficacy
This product is a nicotinic acid derivative. ① Antihypertensive: The exact mechanism of antihypertensive effect is unknown. It mainly dilates arterioles, has little effect on veins, reduces peripheral vascular resistance, increases heart rate, and increases stroke volume and cardiac output. Long-term use can increase renin secretion, aldosterone, and water and sodium retention, thereby reducing the effect. ② Heart failure: This product increases cardiac output and reduces vascular resistance and afterload.

Appearance
This product is white or almost white tablets or sugar-coated tablets.
Indication
Hypertension, heart failure.
Usage and Dosage
The usual oral dose for adults is 10 mg each time, 4 times a day, taken after meals. After 2-4 days, increase to 25 mg 4 times a day for a week; after the second week, increase to 50 mg each time, 4 times a day. The maximum dose does not exceed 300 mg per day. The usual oral dose for children is 750 μg/kg body weight or 25 mg/m2 body surface area, 2-4 times a day, and gradually increase to the maximum dose of 7.5 mg/kg or 300 mg per day within 1-4 weeks.
Adverse Reactions
1 Common: headache, nausea, vomiting, diarrhea, palpitations, tachycardia, etc. 2 Rare: constipation, hypotension, flushing, tearing, nasal congestion. 3 Rare: due to immune allergic reaction, long-term and large-scale use (more than 400 mg/day) can cause rash, itching; chest pain; lymphadenopathy; peripheral neuritis; edema; lupus erythematosus syndrome.
Precautions
Patients with aortic aneurysm, cerebral stroke, and severe renal dysfunction should be considered contraindications.
Special Population Medication
Precautions for children: Not clear yet. Precautions for pregnancy and lactation: This product can pass through the placenta, but there is a lack of research in humans. It is not clear whether it is excreted into breast milk, so it is not recommended for nursing mothers. Precautions for the elderly: The elderly are more sensitive to the antihypertensive effect of this product and are prone to reduced renal function, so it is advisable to reduce the dose.
Drug Interactions
1. When used with nonsteroidal anti-inflammatory analgesics, the antihypertensive effect may be weakened; 2. When used with sympathomimetic amines, the antihypertensive effect of this product may be reduced. 3. When used with diazoxide or other antihypertensive drugs, the antihypertensive effect may be enhanced.
