    Isoluminol, also known as 4-aminophthalic hydrazide, is a white powder with CAS number 3682-14-2. Luminol is a kind of luminol chemiluminescent reagent. Its luminescent mechanism is oxidation reaction luminescence. It is oxidized by H2O2 to form the excited state intermediate of 4-aminophthalic acid under alkaline condition through horseradish peroxidase catalysis. When it returns to the ground state, it will emit photons. It is often used in food, environment, criminal investigation, medicine and other fields. The following will introduce several production methods of isoluminol.

     

    1. Phthalimide was prepared from phthalic anhydride and urea, then 4-nitro-phthalimide was prepared under the catalysis of concentrated sulfuric acid and fuming nitric acid, then 4-nitro-phthalimide was prepared by adding hydrazine hydrate, and then isoluminol was prepared under the catalysis of stannous chloride in alkaline solution. This method has the advantages of simple synthesis process and simple operation. However, when stannous chloride is used as catalyst, a large amount of inorganic tin waste will be generated, which causes great environmental pollution and does not meet the requirements of green synthesis.

     

    2. Diacetyl phthalic hydrazide was prepared by reaction of phthalic hydrazide with acetylation reagent, then reacted with nitroso compound to obtain nitroso diacetyl phthalic hydrazide, then deacetylated to obtain nitroso phthalic hydrazide, and then reduced to isoluminol. This method makes full use of a large number of phthalic hydrazide by-products in the existing synthesis of Gabriel amine as raw materials. The raw materials are cheap and easy to obtain. At the same time, the synthesis process is simple and the yield of isoluminol product is high, but the advantages are also its disadvantages. Using Gabriel amine by-products as raw materials, its quality is not stable and the difference between batches is large. In addition, nitroso compounds are the intermediates in the preparation process It is not suitable for large-scale industrial production.

     

    3. Firstly, phthalimide was prepared, then reacted with concentrated sulfuric acid and fuming nitric acid, cooled and filtered to obtain 4-Nitrophthalimide. Then, the preparation of 4-nitrophthalic hydrazide is mixed with hydrazine hydrate and water of certain concentration and refluxed to obtain orange red solution. After cooling, pH is adjusted with glacial acetic acid, and light yellow solid 4-nitrophthalic hydrazide is obtained by precipitation, washing, filtration and drying. Finally, 4-nitrophthalic hydrazide is thermally dissolved in sodium hydroxide solution, Under constant stirring, the solution is dropped into a pure clear solution containing 56g concentrated hydrochloric acid of stannous chloride, nano titanium dioxide is added, and then it reacts under ultraviolet light to obtain a colorless clear solution, which is cooled, precipitated, filtered, precipitated and filtered, Then the white powdery solid is obtained by drying, which is called isoluminol; this method is cumbersome and not suitable for industrial production, but it has the advantages of high yield and high purity.
