    This drug is a folic acid antagonist, the only difference with folic acid is that the hydrogen on the NH on N10 is changed to methyl (CH3), and the C4 ketone group on the ring is changed to amino (NH2), so it can compete with folic acid for the same enzyme, and it is an antimetabolic therapy drug that can prevent (uracil) from transferring a methyl group through folic acid to form (thymine), which is an antitumour drug, or a drug that prevents transplantation from reacting to the host (rejection). It can suppress bone marrow activity and cause hepatitis. It was used in the 1950s when methotrexate was introduced to replace the more toxic antifolate Aminopterin. It was originally synthesised by Yellapragada Subbarao, an Indian biochemist who spent most of his career in the United States, and clinically developed by Sidney Farber, an American paediatrician. This is a drug on the World Health Organisation's Essential Medicines List, the most important list of medicines that are required in the primary health care system. It is one of the most important anti-rheumatoid arthritis (RA) medications, also known as disease-modifying antirheumatic drugs (DMARDs), because it reduces the inflammatory activity of some of the white blood cells (antibodies), further slowing down the damage to bones.
