Elamipretide, also known as MTP 131, is a mitochondria-targeted peptide antioxidant. Elamipretide decreases inflammation and protect against a variety of neurological illnesses. Elamipretide (SS-31) improves mitochondrial dysfunction, synaptic and memory impairment induced by lipopolysaccharide in mice. Elamipretide Promotes Mitophagosome Formation and Prevents Its Reduction Induced by Nutrient Excess in INS1 β-cells.
Elamipretide Synthetic is a single, non-glycosylated polypeptide chain containing 4 amino acids, having a molecular mass of 640 Dalton and a Molecular formula of C32H49N9O5 .

Mechanism:

A cell-permeable, synthetic tetrapeptide (D-Arg-Dmt-Lys-Phe-NH₂) that selectively concentrates in the inner mitochondrial membrane and reversibly binds to cardiolipin, a phospholipid essential for electron transport chain (ETC) function. By stabilizing cardiolipin, SS-31 restores mitochondrial bioenergetics, reduces cytochrome c peroxidase activity, and prevents oxidative damage in cells with impaired mitochondrial function.

Activity note:

SS-31 does not act via traditional receptor agonism. It binds cardiolipin with high selectivity in the inner mitochondrial membrane. Functional EC₅₀ values are context-dependent (cell type, stress model). Key electrophysiology studies report mitochondrial membrane potential restoration at nanomolar concentrations.
