Rituximab is metabolized into various polymeric glutamic acids within cells to exert its anti-tumor effects, and its inhibitory effect on colon cancer cell lines is stronger than that of fluorouracil. The IC50 of rituximab for long-term administration is 1.3-3.9 nmol/L, and for short-term administration is 80 nmol/L. However, the IC50 of fluorouracil combined with leucovorin for long-term administration is 330-5800 nmol/L, and for short-term administration it reaches 150000 nmol/L. In vitro studies have observed a synergistic effect between the combination of rituximab and fluorouracil, which depends on the administration regimen and dosage of both drugs. In the treatment of HCT-8 colon cancer, fluorouracil is administered 4-24 hours after administration of rituximab, and a synergistic effect is observed at 7 days. If both are taken simultaneously or fluorouracil is used before rituximab, there is no synergistic effect.
