    Description

    Loratadine is a second-generation histamine H1 receptor antagonist used in the treatment of allergic rhinitis and urticaria. Unlike most classical antihistamines it lacks central nervous system depressing effects such as drowsiness. It has been shown to inhibit the release of leukotriene C4 and histamine from rodent mast cells and to inhibit allergic bronchospasm in guinea pigs. Loratadine is a selective inverse peripheral histamine H1-receptor agonist.

    1. Drug Name

    Loratadine is a second - generation anti - histamine drug and a long - acting tricyclic anti - histamine.

    2. Pharmacological Action

    (1) Mechanism of Action

    Loratadine can selectively act on the peripheral H1 - receptors for a long time. It competitively inhibits the binding of histamine to H1 - receptors, thus reducing various allergic reaction symptoms caused by histamine. Histamine is a key mediator in allergic and inflammatory reactions. When the body is exposed to allergens, mast cells and basophils release histamine. After histamine binds to H1 - receptors, it leads to a series of reactions such as vasodilation, increased capillary permeability and smooth muscle contraction, causing itching, redness, sneezing, runny nose and other allergic symptoms. Loratadine alleviates allergic reactions by blocking this process.

    (2) Efficacy Characteristics

    It has the characteristics of quick onset. Generally, it can take effect 1 - 3 hours after oral administration. Moreover, its efficacy lasts for a long time, about 24 hours, which enables patients usually to take the drug only once a day to effectively control the symptoms.
