Application.
Nonionic detergent.

TWEEN 20 is a Nonionic detergent widely used in biochemical applications. It has been used as an emulsifier to prepare stable oil-in-water emulsion. TWEEN 20 has been used to pre-extract cell membranes to remove peripheral proteins (membrane-binding proteins can be extracted at 2% concentration). TWEEN 20 has been used as a blocking agent for membrane-based immunoassays with a typical concentration of 0.05%. TWEEN 20 can be used to dissolve mammalian cells at concentrations of 0.005 to 0.5%.

Warning.
High pressure sterilization of TWEEN 20 may be unstable, especially when there are metal ions in the buffer solution. High pressure sterilization is not recommended when the characteristic changes have not been determined. TWEEN 20 is sensitive to heat and darkens when exposed to high temperatures. It is reported that polysorbate is incompatible with alkali, heavy metal salts, phenols and tannic acids. They may also reduce the activity of many preservatives. The aqueous solution of polysorbate will self-oxidize during storage, and its change is catalyzed by light, rising temperature and copper sulfate.

Points for attention in preparation.
TWEEN 20 is soluble in water (100mg / mL) and produces a clear yellow solution. It is also soluble in ethanol, dioxane and ethyl acetate. TWEEN 20 is insoluble in liquid paraffin and fixed oil.
