Introduction
Sorafenib (trade name: Doruxime) is a new molecular targeted therapy drug. It is an oral multi-target and multi-kinase inhibitor that can target serine/threonine kinases and receptor tyrosine kinases on tumor cells and tumor blood vessels. Sorafenib can inhibit receptor tyrosine kinases such as KIT and FLT-3, as well as Raf/MEK/ERK pathway serine/threonine kinases, significantly inhibiting tumor cell proliferation. On the other hand, by upstreamly inhibiting receptor tyrosine kinases VEGFR and PDG-FR, and downstreamly inhibiting serine/threonine kinases in the Raf/MEK/ERK pathway, it significantly inhibits tumor angiogenesis. That is, it can achieve dual effects by killing two birds with one stone, exerting dual functions of anti-angiogenesis and anti-tumor cell proliferation simultaneously.

Pharmacological Effects
Sorafenib is a multi-kinase inhibitor. Preclinical studies have shown that sorafenib can simultaneously inhibit various kinases present both in the intracellular and on the cell surface, including RAF kinases, vascular endothelial growth factor receptor-2 (VEGF-2), vascular endothelial growth factor receptor-3 (VEGF-3), platelet-derived growth factor receptor-β (PDGFChemicalbookR-β), KIT, and FLT-3. Thus, sorafenib exhibits dual anti-tumor effects. On one hand, it can directly inhibit tumor growth by suppressing the RAF/MEK/ERK signaling pathway; on the other hand, it can also block the formation of tumor new blood vessels by inhibiting VEGFR and PDGFR, thereby indirectly inhibiting the growth of tumor cells.
