Description	Tacrolimus, also known as FK-506, is an immunosuppressive drug used mainly after allogeneic organ transplant to reduce the activity of the patient's immune system and to lower the risk of organ rejection. It is also used in a topical preparation in the treatment of atopic dermatitis (eczema), severe refractory uveitis after bone marrow transplants, exacerbations of minimal change disease, TH2-mediated diseases such as Kimura's disease, and the skin condition vitiligo. FK-506 is a macrolide isolated from the fungus Streptomyces tsukubaensis. Note: this product is replaced by tacrolimus hydrate.

Tacrolimus More Info

Tacrolimus, a macrolide immunosuppressive agent, is widely used to prevent organ rejection after transplantation. It functions by suppressing the immune system's response to foreign tissues, thus preventing the body from attacking the transplanted organ.

Mechanism of Action:

Tacrolimus works by inhibiting calcineurin, an enzyme involved in the activation of T-cells. By blocking calcineurin, tacrolimus prevents the production of interleukin-2 (IL-2), a cytokine that promotes the proliferation T of-cells. This action leads to a reduction in the immune response against the transplanted tissue, thereby reducing the risk of organ rejection.

Indication:

Tacrolimus is primarily indicated for the prophylaxis of organ rejection in patients receiving kidney, liver, heart, lung, or pancreas transplants. It is also used in the treatment of severe autoimmune diseases such as psoriasis, rheumatoid arthritis, and uveitis when other treatments have been ineffective.

Pharmacodynamics:

Tacrolimus is most effective when taken consistently as part of a comprehensive immunosuppressive regimen that may include other drugs like corticosteroids and anti-metabolites. It is typically administered orally, and blood levels are closely monitored to ensure therapeutic efficacy while minimizing side effects.
