    Discovery and Development

    TFFH was first introduced by Carpino and El-Faham in 1995 as a safe and user-friendly alternative to corrosive cyanuric fluoride, initially for peptide synthesis applications.

    Key Applications

    Peptide Synthesis: It serves as an excellent coupling reagent for both solution and solid-phase peptide synthesis, especially for sterically hindered amino acids (e.g., Aib-containing peptides). In the presence of DIPEA, it converts Fmoc-amino acids to acid fluorides (1842 cm⁻¹ IR absorption) for efficient amide bond formation.

    Organic Synthesis: It is used to prepare acid fluorides, hydrazides, azides, anilides, alcohols, and hydroxamic acids from carboxylic acids. Combined with benzyltriphenylphosphonium dihydrogen trifluoride (PTF), it converts carboxylic acids to alcohols with high yields.

    Deoxofluorination: It enables deoxofluorination reactions, which can be scaled up to over 100 grams, suitable for industrial applications.

    Handling and Advantages

    Handling: It is non-hygroscopic and air-stable, allowing routine handling without special precautions. In automated peptide synthesis, it is dissolved in DMF with Fmoc-amino acid and DIPEA, agitated for 7 minutes to form acid fluoride before resin coupling.

    Advantages: Compared to other fluorinating reagents (e.g., XtalFluor-E/M), it has lower exothermic heat during decomposition, enhancing safety. It avoids the formation of symmetric anhydrides when used with PTF, improving product purity and yield.

    Safety Considerations

    It decomposes at 370°C, releasing heat, so avoid exposure to high temperatures.

    As a fluorinated compound, it should be handled in a well-ventilated fume hood, with appropriate PPE (gloves, goggles, lab coat).

    Store in a cool, dry place away from incompatible substances (e.g., strong oxidizers, strong bases).
