Application&Function

Setipiprant functions primarily as a selective antagonist of the CRTH2 receptor, a G-protein-coupled receptor expressed on Th2 lymphocytes, eosinophils, and basophils. By binding competitively to CRTH2, it potently blocks the activation signals induced by prostaglandin D2 (PGD2), a key lipid mediator involved in allergic and inflammatory pathways. This inhibition prevents PGD2-induced chemotaxis of Th2 cells and eosinophils, reduces the release of pro-inflammatory cytokines (such as IL-4, IL-5, and IL-13), and suppresses eosinophil degranulation.

Therapeutically, Setipiprant has been investigated primarily for the treatment of allergic and inflammatory conditions, including asthma, allergic rhinitis, and atopic dermatitis. By interrupting the PGD2-CRTH2 axis, it attenuates the inflammatory cascade characteristic of type 2 immunity, thereby reducing airway inflammation, bronchoconstriction, and symptoms of allergic response. Its oral bioavailability and receptor selectivity offer a targeted mechanism with potential advantages over broader anti-inflammatory agents.
