    1. Description

    1,3-Dihydroxyacetone is the simplest ketose (a three-carbon sugar) and belongs to the class of polyhydroxy ketose compounds. It appears as white crystalline powder with a sweet taste, has a melting point of 75–80°C, and is readily soluble in water, ethanol, diethyl ether, acetone, and other organic solvents. Its molecular formula is C₃H₆O₃, and its molecular weight is approximately 90.08 Da. In the solid state, this compound primarily exists as a dimer; upon dissolution, it dissociates into a mixture of monomers and diol hydrates. It must be stored in a sealed container, protected from light, and refrigerated at 4°C, as it is hygroscopic.

    2. Application

    (1) Cosmetics and Skincare: As the key active ingredient in self-tanning products, it is added to sunscreens, lotions, sprays, and other products. Its ketone functional group undergoes the Maillard reaction with the amino groups of keratin in the skin’s stratum corneum, producing brown polymers (melanin-like compounds), thereby mimicking the skin tone resulting from sun exposure. According to EU cosmetics regulations, the maximum permitted concentration in tanning products is 10%, and 6.25% in oxygen-free hair dye products.

    (2) Functional Foods and Nutritional Supplements: Used as a food additive, it has the potential to promote energy metabolism and fat burning. Studies indicate that it can increase metabolic rate and fatty acid oxidation, making it valuable for applications in the functional food sector.

    (3) Antitumor Research: In an H460 human large-cell lung cancer xenograft model, the combination of dihydroxyacetone and cisplatin demonstrated a synergistic effect, with a tumor weight inhibition rate of 53.3%, outperforming the cisplatin-only group (44.6%) without increasing cisplatin-related toxic side effects. Its antitumor mechanism involves inducing apoptosis and regulating redox status.

    (4) Organic Synthesis Intermediates: As an important precursor for synthesizing various fine chemicals, it has wide applications in the fields of pharmaceutical and pesticide synthesis. It also serves as a specific biochemical substrate for galactose oxidase.

    3. Main Efficacy

    (1) Skin tanning effect: Through the Maillard reaction, it binds with free amino groups in skin keratin (particularly the guanidino group of arginine) to form brown polymers. This reaction is confined to the stratum corneum and does not penetrate into living cells; the resulting tan can last for 3–10 days. Compared to UV-induced tanning, it is a relatively safe alternative that helps avoid the risks of skin cancer, sunburn, and photoaging caused by UV radiation.

    (2) Promotion of Fat Metabolism and Energy Expenditure: Animal studies indicate that dihydroxyacetone can increase metabolic rate and fatty acid oxidation, effectively burning fat and reducing body fat, thereby offering potential weight-loss benefits. Long-term supplementation can enhance glucose utilization and conserve muscle glycogen, which is beneficial for improving exercise endurance.

    (3) Antitumor Activity: In an H460 lung cancer nude mouse model, the combination of dihydroxyacetone and cisplatin significantly enhanced antitumor effects, reducing the tumor proliferation rate (T/C) to 44.9% and achieving a 53.3% tumor weight suppression rate. Its mechanism of action may involve inducing cell cycle alterations, regulating redox status, and enhancing sensitivity to chemotherapeutic agents.

    (4) Antioxidant and Anti-inflammatory Potential: Although the Maillard reaction produces small amounts of reactive oxygen species (ROS), modern formulations often include natural antioxidants to mitigate potential oxidative damage. At appropriate concentrations, dihydroxyacetone exerts a regulatory effect on cellular oxidative stress.

    (5) Regulation of Glucose Metabolism: As an intermediate in glucose metabolism, it participates in the body’s energy metabolism pathways. Studies indicate that it can regulate cAMP levels and influence the activity of various metabolic enzymes.
