(1R,2R)-1,2-Cyclohexanedicarboxylic acid is the key intermediate of rulaxitide hydrochloride. It can be obtained by the resolution of trans-1,2-cyclohexanedicarboxylic acid. Rulaxitide hydrochloride is a novel atypical antipsychotic drug developed by Sumitomo Pharmaceutical Company of Japan. It was first launched in the United States in 2010 and was approved by the FDA for the treatment of adult schizophrenia and as a monotherapy or as an adjunct therapy with lithium salts or valproic acid for depressive episodes associated with bipolar I disorder in adult patients. It was approved by the EMA in 2014 for marketing in the European Union. It has a high binding affinity for dopamine D2 receptors and 5-hydroxytryptamine (5-HT1A, 5-HT2A, 5-HT7) and α2C receptors, with relatively low binding affinity for other receptors. It has significant therapeutic effects on both positive and negative symptoms of patients with mental disorders. Studies have reported that rulaxitide can improve cognitive function, cause less weight gain, and not cause changes in glucose, lipids (lipids), ECG, or QT interval.
