As a direct upgrade and replacement for Benzotriazol-1-yl-oxytris(dimethylamino)phosphonium hexafluorophosphate (BOP), PyBOP represents a milestone in the field of peptide synthesis.

A Safe and Environmentally Friendly “Green” Alternative: This is PyBOP’s most significant advantage. The classic condensation reagent BOP inevitably generates the byproduct hexamethylphosphamide (HMPA) during the reaction; this substance has been proven to be carcinogenic and is listed on the “Seveso Directive” list of hazardous chemicals. By replacing the dimethylamino group in the BOP structure with a pyrrolidyl group, PyBOP completely eliminates the formation of HMPA, making the synthesis process safer and more environmentally friendly, in line with the principles of green chemistry.

High Coupling Efficiency: PyBOP not only serves as an excellent substitute for BOP but also maintains equivalent or even superior reactivity. It can rapidly initiate reactions even when encountering amino acids with significant steric hindrance. Studies have shown that the degree of amino acid racemization is reduced by approximately 50% when using PyBOP compared to BOP, which is crucial for ensuring the optical purity and bioactivity of peptide products.

Minimal side reactions: Unlike uronium-based condensation reagents, PyBOP does not induce guanidination of amino groups as a side reaction during the reaction. Physical Properties: Typically a white to off-white fine crystalline powder with a melting point between 140–160°C. Purity is typically required to be 98% or higher (HPLC).

Storage Conditions: Highly hygroscopic; it is recommended to store in a sealed container in a cool, dry environment at 2–8°C. For long-term storage, it is recommended to store at -20°C and ensure it is kept dry under an inert gas atmosphere to prevent degradation.
