Product Overview

Benzotriazol-1-yloxytris(dimethylamino)phosphonium hexafluorophosphate (BOP, CAS 56602-33-6) is a peptide coupling reagent with the molecular formula C₁₂H₂₂F₆N₆OP₂ and a molecular weight of 442.28 g/mol. The compound is widely used in peptide synthesis for the activation of carboxylic acids to form amide bonds. It is a white to off-white crystalline powder with good solubility in organic solvents such as DMF and DCM.

Synthetic Process

BOP is synthesized by the reaction of benzotriazole with tris(dimethylamino)phosphonium chloride followed by anion exchange with hexafluorophosphate.

Mechanism of Action

BOP activates carboxylic acids by forming a reactive O-benzotriazolyl ester intermediate, which is then attacked by an amine to form the amide bond. It is particularly useful for the synthesis of peptides and peptidomimetics.
