    • Halting the Quality Control System: Hsp90 normally acts as a quality control chaperone, helping mutated or unstable oncogenic proteins fold correctly to prevent them from breaking down. Ubiquitin-Mediated Shredding: By blocking Hsp90's ATPase activity, 17-DMAG stops this protective folding. Unprotected client proteins become misfolded and are rapidly degraded via the ubiquitin-proteasome pathway.
    2. Targeting Multiple Oncogenic Pathways
    Because Hsp90 protects a wide variety of "client proteins" that drive cancer, 17-DMAG acts as a multi-targeted antineoplastic agent. It effectively downregulates: 
    • Pro-Survival Kinases & Receptors: Destabilizes proteins like MET, Akt, HER2, and BCR-ABL.
    • Transcription and Cell Cycle Regulators: Suppresses critical cancer drivers, including NF-κB, Cyclin D1, and Survivin, leading to G2/M phase cell cycle arrest and early apoptosis (programmed cell death).
    • ROS Generation: Induces therapeutic stress in cancer cells by accelerating the production of Reactive Oxygen Species (ROS) while simultaneously suppressing their internal antioxidant enzymes.
    3. Unique Molecular Target: NF-κB and IKK Depletion
