In vivo studies in animal models show that Valerenic acid produces anxiolytic, sedative, and sleep-promoting effects, with influences on neuronal excitability, stress responses, and behavioral regulation. 2. Mechanism of Action

Valerenic acid is an orally active positive allosteric modulator of GABAA receptors and a partial agonist of the 5-HT5a receptor. Key mechanistic features include:

GABAA receptor modulation: It displays a preference for β2 or β3 subunit-containing GABAA receptors. It directly activates the receptor and blocks the channel at high concentrations. Its anxiolytic action is mediated through GABAA receptors containing the β 3 subunit.

5-HT5a receptor: Acts as a partial agonist of the 5-HT5a receptor.

Antioxidant activity: Possesses potent antioxidant properties.
