N-Acetyl-L-cysteine ethyl ester (CAS 59587-09-6), also known as N-Acetylcysteine ethyl ester or NACET, is a lipophilic, cell-permeable derivative of N-acetylcysteine (NAC). With a molecular formula of C₇H₁₃NO₃S and a molecular weight of 191.25 g/mol, it is designed to overcome the poor bioavailability of NAC by esterifying its carboxyl group, which drastically increases lipophilicity.

Once inside cells, NACET efficiently releases NAC and cysteine, serving as an effective precursor for glutathione (GSH) synthesis. It acts as a mucolytic agent, a paracetamol antidote, and a GSH-related antioxidant that protects against hydroperoxide-induced oxidative damage. NACET also increases circulating hydrogen sulfide (H₂S), making it a promising oral H₂S donor. Studies have shown that it enhances intracellular glutathione concentrations and protects cells from oxidative stress. With significantly improved bioavailability compared to NAC, NACET has the potential to substitute NAC in various therapeutic and research applications
