    DSIP, or delta-sleep-inducing peptide, is a synthetic nonapeptide molecule with the following sequence. This peptide was initially isolated from animal samples. Extensive research has focused on its regulatory effects in vitro and in animal models, including sleep control, stress regulation, and endocrine intervention. As a research-grade peptide raw material, DSIP is widely used in peptide laboratory research, sleep pharmacology, and endocrinology-related projects, and is one of the most studied peptides in the field of research raw materials internationally.

    DSIP consists of nine short-chain peptides. Under natural conditions, this molecule exists in trace amounts in the central nervous system. High-purity DSIP powder, typically exceeding 98%, can be prepared using artificial solid-phase peptide synthesis technology; it is usually a white lyophilized powder. This powder exhibits good stability under low-temperature, light-protected storage conditions, but its stability in aqueous solutions is relatively poor. Therefore, in research, DSIP is usually used immediately after preparation. DSIP cannot directly cross the skin barrier; therefore, targeted drug delivery is typically employed in research models, a key physicochemical characteristic for the scientific application of this peptide.

    One of its core research directions is the regulation of delta-wave sleep, hence the name DSIP (Digital Subtraction Angiogenesis-Induced Sleep). Sleep is divided into rapid eye movement (REM) sleep and non-rapid eye movement (NREM) sleep. Delta waves are the hallmark electrical waves of deep sleep, while deep slow-wave sleep is associated with bodily repair, memory consolidation, and the release of circadian rhythms. Numerous animal studies have shown that DSIP intervention can alter the proportion of delta waves in experimental animals, thereby affecting the duration of nighttime slow-wave sleep. Its mechanism of action is not merely to induce drowsiness, but focuses on regulating the sleep-wake cycle and optimizing sleep structure. Unlike commonly used sedatives, DSIP research models rarely show a significant decline in arousal inhibition or motor function; instead, it primarily reconstructs the sleep-wake rhythm, demonstrating the potential to regulate circadian rhythm disorders. In animal models of jet lag and circadian rhythm reversal, DSIP has been used to study resetting the biological clock and improving sleep fragmentation, providing a molecular tool for basic research on circadian rhythm disorders.

    The second major research area is stress and its regulation. When the body is under chronic stress, persistent activation leads to abnormally elevated levels of cortical stress hormones, which in turn interfere with sleep, immunity, and mood. Animal experiments have shown that DSIP can modulate the hypothalamic-pituitary-adrenal (HPA) axis, inhibit excessive stress signal transduction, and downregulate abnormal stress release under stress. In animal models of physiological and psychological environmental stress, the DSIP treatment group showed a trend of improvement in anxiety-related behavioral indicators. From a research perspective, DSIP does not directly block stress perception, but rather helps the body recover from a state of sustained high stress to homeostasis by balancing overactive stress feedback loops through central nervous pathways. Much basic research in this field revolves around neurotransmitter interactions. DSIP interacts with multiple central neurotransmitter systems, indirectly regulating inhibitory balance. This is the molecular mechanism by which it alleviates stress-related symptoms.

    Endocrine regulation is another important research direction for DSIP. The pulsatile release of growth hormone is highly dependent on deep slow-wave sleep, with its secretion peak occurring during deep sleep at night. When sleep structure is disrupted and delta sleep is reduced, the pulsatile release of growth hormone is also inhibited. DSIP indirectly affects the circadian rhythm of growth hormone secretion by regulating delta-slow-wave sleep, and changes in growth hormone release levels can be observed in animal models. Simultaneously, this peptide can also regulate adrenocorticotropic hormone (ACTH), thereby affecting the function of the hypothalamus-pituitary axis.
