SS-31 (Elamipretide, Bendavia, MTP-131) is a synthetic tetrapeptide with the sequence D-Arg-Dmt-Lys-Phe-NH2, developed in the laboratory of Dr. Hazel Szeto at Cornell University research College. It is a cell-permeable, mitochondria-targeted peptide that selectively binds to cardiolipin, a phospholipid unique to the inner mitochondrial membrane. By binding cardiolipin, SS-31 stabilizes mitochondrial membranes, reduces mitochondrial reactive oxygen species (ROS) production, and preserves mitochondrial function during oxidative stress. SS-31 has been investigated in research studys for heart failure, mitochondrial myopathy, and rare mitochondrial diseases.

As a research reagent, SS-31 is intended for in vitro and in vivo studies of mitochondrial function, oxidative stress, cardioprotection, neuroprotection, renal protection, and aging. Research has shown that SS-31 can reduce mitochondrial ROS, preserve ATP production, inhibit mitochondrial permeability transition pore opening, and protect cells from oxidative damage in various disease models.

• Cardiolipin Binding :SS-31 selectively binds to cardiolipin in the inner mitochondrial membrane through electrostatic and hydrophobic interactions. This binding stabilizes the mitochondrial membrane structure, preserves cristae morphology, and maintains the proper spatial organization of the electron transport chain complexes.
• Mitochondrial ROS Reduction ：By stabilizing the electron transport chain and reducing electron leakage, SS-31 decreases mitochondrial superoxide and ROS production. Unlike conventional antioxidants that scavenge ROS after they are formed, SS-31 prevents ROS production at the source by improving mitochondrial electron transport efficiency
• ATP Production Preservation ：SS-31 preserves mitochondrial ATP production during oxidative stress by maintaining mitochondrial membrane potential and electron transport chain function. Studies have shown that SS-31 can improve ATP levels in tissues with mitochondrial dysfunction, including heart, brain, kidney, and skeletal muscle.
• mPTP Inhibition ：SS-31 inhibits the opening of the mitochondrial permeability transition pore (mPTP), a key event in mitochondria-mediated apoptosis and cell death.
