Trilaciclib dihydrochloride (CAS 1977495-97-8), also known as G1T28 dihydrochloride, is an orally bioavailable, potent and selective inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6) with the molecular formula C₂₄H₃₂Cl₂N₈O and a molecular weight of 519.48 g/mol . The compound is a pyrimidine-benzimidazole derivative featuring a 4-methylpiperazin-1-yl pyridin-2-yl amino substituent and a spiro[cyclohexane-1,9'-pyrazino[1',2':1,5]pyrrolo[2,3-d]pyrimidin]-6'-one core . Trilaciclib was approved by the FDA on February 12, 2021, and is indicated to reduce the incidence of chemotherapy-induced myelosuppression in adult patients with extensive-stage small cell lung cancer receiving platinum/etoposide or topotecan-containing regimens
