5-Pyrimidinemethanol Use and Manufacturing

Step 3. Pyrimidin-5-ylmethanol. A mixture of pyrimidine-5-carboxaldehyde (2 g, 18.50 mmol, 1.00 equiv) and sodium borohydride (2 g) in methanol (100 mL) was stirred at 0- 10°C for 30 mm. The reaction mixture was concentrated under vacuum and the residue was purified on a silica gel column eluted with dichloromethane/methanol (50:1) to yield 1.2 g (59percent) of pyrimidin-5-ylmethanol as a light yellow solid. MS: m/z = 111.0 [M+H].To a solution of pyrimidine-5-carbaldehyde (0.300 g, 2.77 mmol) in methanol (6 mL), sodium borohydride (0.158g, 4.16 mmol) was added at 0 °C over 10 minutes and the reaction mixture was stirred at the same temperature for 30 min. After completion of the reaction, the solvent was removed under vacuum. The residue obtained was poured into water (30 mL), extracted with ethyl acetate (150 mL), The organic layer was dried over sodium sulfate and concentrated to provide the title compound (0.160 g, 52.4percent), A mixture of pyrimidine-5-carboxaldehyde (2 g. 1 8.50 mmol, 1 .00 equiv) and sodium borohydnde (2 g) in methanol ( 100 mL) was stirred at 0-10°C for 30 min. The reaction mixture was concentrated under vacuum and the residue was purified on a silica gel column eluted with dichloromethane/methanol (50:1) to yield 1.2 g (59percent) of pyrimidin-5-ylmethanol (commercially available, C AS 251 93-95-7) as a light yellow solid. LC/MS (Method N ESI): RT= 0.74 min, m/z = 111.0 [M+H]+A mixture of pyrimidine-5-carboxaldehyde (2 g. 1 8.50 mmol, 1 .00 equiv) and sodium borohydnde (2 g) in methanol ( 1 00 m L ) was stirred at 0 - 1 0°C for 30 min. The reaction mixture was concentrated under vacuum and the residue was purified on a silica gel column eluted with dichloromethane/methanol (50: 1 ) to yield 1 .2 g (59percent) of pyrimidin-5-ymethanol (commercially available, C AS 251 93-95-7) as a light yellow solid. LC MS (Method N ESI): RT= 0.74 min, m/z = 111.0 [M+H]+A mixture of pyrimidine-5-carboxaldehyde (2 g, 1 8.50 mmol, 1 .00 equiv) and sodium borohydride (2 g) in MeOH (100 mL) was stirred at 0 - 10°C for 30 min. The reaction mixture was concentrated under vacuum and the residue was purified on a silica gel column eluted with DCM/MeOH (50: 1 ) to yield 1 .2 g (59percent) of pyrimidin-5-ylmethanol as a light yellow solid. LC/MS (Method K, ESI): RT= 0.74 min, m z = 1 1 1 0 [M+H]0226] Step 3. Pyrimidin-5-ylmethanol.
