4,5-Dichloro-2-fluorobenzoic acid Use and Manufacturing

To a stirred solution of (Int-IG) (26 mg, 0.12 mmol, 1 equiv) in dry DMF (2 mL) was added EDCI (35 mg, 0.18 mmol, 1.5 equiv), HOBt (24 mg, 0.18 mmol, 1.5 equiv), To To a solution of 4, 5-dichloro-2-fluoro-benzoic acid (5.0 g, 24 mmol) in tetrahydrofuran (50 mL) at 0 C was added borane -tetrahydrofuran complex (1 M, 60 mL, 60 mmol) dropwise over 30 minutes. The resulting solution was stirred at 30 C for 14 hours until TLC analysis showed the reaction was complete. The reaction was quenched carefully with methanol (10 mL) and then concentrated in vacuo to give compound A-21 (4.50 g, 85% yield) as a yellow oil, which was used in the next step without further purification. LCMS (DD): (ES+) m/z (M+H)+ = 176.9, tR=0.859 min.
