Eprodisate Disodium is the orally available disodium salt form of Eprodisate, a negatively charged sulfonated inhibitor of fibrillogenesis, that can be used in the treatment of amyloid A (AA) amyloidosis.
Eprodisate Disodium is the orally available disodium salt form of eprodisate, a negatively charged sulfonated inhibitor of fibrillogenesis, that can be used in the treatment of amyloid A (AA) amyloidosis. Upon administration, eprodisate competitively binds to the glycosaminoglycan binding sites on serum amyloid A (SAA), which inhibits the formation of the glycosaminoglycan-amyloid fibril aggregate. This prevents the formation of amyloid deposits in certain organs, especially the kidneys, in AA amyloidosis.

To a suspension of disulfone from Step 2 (0.62 g, 2.23 mmol) in methanol (15 mL) was added a 0.5M solution of sodium methoxide in methanol (8.9 mL, 4.58 mmol). After 24 h of stirring, the homogenous solution was concentrated in vacuo and the resulting solid was suspended in ethanol (20 mL) and the suspension was stirred 1 h at room temperature. The suspension was filtered and the cake washed with ethanol (2x10 mL) and diethyl ether (2x10 mL). Water (2 mL) was added to the solid and the mixture passed through a C8 pad eluting with water. The fractions containing Compound P1(disodium salt) were collected and lyophilized, affording Compound P1(disodium salt) (0.41 g, 85percent yield) as a white solid.
