4-amino-6-chloronicotinaldehyde Use and Manufacturing

Scheme 8 compound 4 (600 mg, 3.77 mmol) was dissolved in THF (50 mL). To this solution was added Mn0To a solution of (4-amino-6-chloro-pyridin-3-yl)-methanol (0.335 g, 2.1 mmol) (from Example 23 supra) in dichloromethane (100 mL) at room temperature was added MnOTreat a solution of (4-amino-6-chloropyridin-3-yl)methanol (10 g, 63.3 mmol) in DCM (150 mL) with activated manganese dioxide (38 g, 443 mmol) and stir at RT overnight. Remove solids via filtration, concentrate the filtrate and purify by silica gel chromatography to afford the title compound (7.2 g, 73percent). 1H NMR (400 MHz, DMSO- d(4-Amino-6-chloropyridin-3-yl)methanol (2.6 g, 16.4 mmol) was dissolved in DCM (80 mL) along with Mn02 (14.3 g). The mixture was stirred at rt for 24 h. Mn02 was filtered off andthe filtrate was concentrated to afford 4-amino-6-chloronicotinaldehyde (1.67 g, 65percent) as a white solid.In a dry 250 mL round bottom flask, Add In a dry 250 mL round bottom flask, Step 1.
