5-Amino-1-methylpyrazole-4-carboxamide Use and Manufacturing

A mixture of 5 -amino- 1 -methyl- lH-pyrazolo 4-carbonitrile 2 (13 g, 0.11 mol) in cone, sulfuric acid (30 mL, 0.6 mol) was stirred for Ih at room temperature. The mixture was poured on ice and the resulting mixture was basified to pη = 8 with 50percent ammonia hydroxide aqueous solution in the ice bath. The precipitated product was filtered and washed with water to give 5-amino-l -methyl- lH-pyrazolo-4-carboxylic acid amide 3 (13.5g, 91percent).Alternatively, a mixture of 5 -amino- 1 -methyl- lH-pyrazolo 4-carbonitrile 2 (3.00 g, 24.59 mmol), IM aqueous solution of sodium hydroxide (1.1 equiv., 27 ml) and -30percent aqueous solution of hydrogen peroxide (5 equiv., 14 ml) were stirred in methanol (30 ml) at room temperature overnight. The volume of reaction mixture was reduced in vacuo, precipitate filtered and dried to give 5 -amino- 1 -methyl- lH-pyrazolo-4-carboxylic acid amide 3 (2.99 g, 87 percent).
