6-Bromo-2-methyl-2H-indazole Use and Manufacturing

Part A: To 6-bromoindazole (402A) (5.0 g, 25.4 mmol) in THF (50 mL) was added sodium hydride (95percent, 672 mg, 26.6 mmol) with ice bath cooling. The mixture was stirred for 30 minutes. Methyl iodide (6.36 mL, 102 mmol) was added at room temperature. The reaction mixture was quenched with ammonium chloride solution and the layers were separated. The aqueous layer was extracted with ethyl acetate. The combined organic layer was washed with brine, dried over sodium sulfate and concentrated. Purification by column chromatography (SiOExample 2005 Compound 2005A (400 mg, 2.03 mmol) was dissolved in 10 ml of THF and reacted with sodium methoxide (4 mL, 2.03 mmol). After 30 min iodomethane (0.5 mL, 8.12 mmol) was added. Reaction mixture was stirred for 3 hours at room temp, then it was added to ethyl acetate and water. The aqueous layer was extracted with ethyl acetate twice. All the organic layers were combined, washed with brine solution and dried over sodium sulfate. The solvent was removed by rotary evaporator and further purified by S1OStep 2: Preparation of ethyl 5-(2-methyl-2H-indazol-6-yl)isoxazole-3-carboxylate To a solution of Step 1: Preparation of General procedure: Indole-6-boronic acid (274 mg, 1.7 mmol) and 7a (516 mg, 1.5 mmol)were added into a 100 mL round-bottom flask containing 1, 4-dioxane (10 mL), then 25.2 mgPd(PPh3)2Cl2 was added, followed by 1.0 mL 2.0 M K2CO3. The mixture was stirred and heatedto reflux under Argon for about 4 hours. After the reaction was complete, the mixture was cooledto room temperature, 10.0 mL H2O was added and the product was extracted with ethyl acetate(10 mL×3). The organic solvent was combined, dried (anhydrous Na2SO4), filtered andevaporated. The crude product was purified by chromatographic column using hexane : ethylacetate (4:1) as eluent. 380.1 mg methyl 3-[[6-(1H-indol-6-yl)indol-1-yl]methyl]benzoate wasobtained as pale yellow oil, yield 67%.
